由于APC-R试验是在加入外源性APC条件下进行APTT测定,除FV Leiden突变外,蛋白S缺陷、狼疮抗凝物存在、FⅡ、FⅧ、FⅩ缺乏、口服抗凝药物治疗等均可影响试验结果,急性血栓形成或妊娠妇女由于体内止凝血系统的异常变化也可影响试验结果。为克服上述试验不足,有一种改进方法检测APC-R,方法是将待测血浆用乏FⅤ的血浆稀释后,再加入组织因子、Ca2+和活化蛋白C(APC),测定其加或不加APC的凝血酶原时间(PT)并求出其比值,可更灵敏地判断APC-R,此法对FV Leiden突变检测的灵敏度和特异性均可达到100%。\\
